Description

Department Overview This position requires a PhD in Developmental Biology, Genetics and Preimplantation Embryo Development, Pluripotent Stem Cell Biology, Genome Editing, or a closely related field. The successful candidate will conduct research focused on correction of various pathogenic mutations in gametes and/or early preimplantation embryos using advanced genome editing approaches. The work will involve advanced gamete and embryo manipulation techniques combined with genetic, epigenetic, transcriptomic, genome editing, sequencing, and bioinformatics approaches in support of the mission and strategic initiatives of the OHSU Center for Embryonic Cell and Gene Therapy (CECGT). Function/Duties of Position The candidate will design and execute independent, hands-on experiments; maintain rigorous laboratory documentation; collect, organize, analyze, and interpret complex datasets; and prepare manuscripts for publication. Additional responsibilities include presenting findings at seminars and scientific conferences, developing experimental protocols, contributing to grant applications and progress reports, and preparing technical reports. The position also involves training and mentoring junior personnel. The role requires scientific independence, strong analytical skills, flexibility, and demonstrated expertise in developmental and stem cell biology. Technical Research Responsibilities Independently conduct cutting-edge research focused on the development, optimization, and delivery of gene correction constructs using both in vitro and in vivo model systems. Responsibilities include: Designing, testing and optimizing various gene editing constructs for precise, targeted correction of mutations in induced pluripotent stem cells (iPSCs) and performing comprehensive on-target an off-target evaluations to ensure genomic integrity Targeted correction of mutations in gametes and preimplantation embryos and performing comprehensive on-target an off-target evaluations in gene corrected embryos and ES cells Routine maintenance and quality control of primary and stem cell cultures. Performing comprehensive sequencing and differentiation analyses to assess the functional consequences of genome editing in gene corrected stem cells and tissues. Applying directed differentiation protocols to generate neuronal lineages from iPSCs for phenotypic analysis of genome-edited cells. Data Collection, Analysis, and Record Keeping Collect, analyze, and interpret scientific data relevant to ongoing research projects. Maintain comprehensive, accurate, and well-organized experimental records in accordance with institutional and regulatory standards. Ensure that all research data, protocols, and related documentation are systematically archived and readily accessible for current reference and long-term documentation. Report Preparation and Literature Research Prepare analytical, descriptive, and evaluative reports for publication in peer-reviewed journals and for inclusion in grant applications, progress reports, and presentations at national and international scientific meetings. Conduct comprehensive literature reviews and database analyses to inform experimental design, data interpretation, and strategic research planning. Other Duties Provide hands-on training in experimental techniques and established protocols. Delegate responsibilities appropriately. Oversee the work of students, trainees, and support staff to ensure high standards of scientific rigor, productivity, and compliance. Present research findings to fellow investigators and collaborative scientists; attend professional meetings. Perform additional tasks as required or assigned, to fulfill the mission of the Center for Embryonic Cell and Gene Therapy. Required Qualifications Ph.D. in Developmental Biology, Stem Cell Biology, or a closely related field. Demonstrated expertise in designing and implementing genome editing strategies, including CRISPR/Cas9, base editing, and prime editing, for the correction of pathogenic mutations in mammalian gametes, embryos, and induced pluripotent stem cells (iPSCs). Proven experience in comprehensive on-target and off-target analyses of gene editing outcomes in gametes, embryos, and embryonic stem cells (ESCs) using advanced sequencing technologies and bioinformatics approaches. Excellent written and verbal communication skills, with a strong record of first-author publications in peer-reviewed journals on related topics. Demonstrated ability to think strategically and execute research plans efficiently within defined timelines. Strong analytical skills, with the ability to critically analyze, integrate, and interpret complex datasets. Ability to work independently as well as collaboratively within small, interdisciplinary teams. Proven ability to interact professionally and productively with diverse staff and trainees. Strong organizational skills, attention to detail, and the ability to prioritize competing responsibilities while coordinating research activities effectively. Preferred Qualifications Demonstrated hands-on expertise in in vitro mammalian gamete and embryo technologies, including in vitro fertilization (IVF), somatic cell nuclear transfer (SCNT), mitochondrial replacement techniques (MRT), and embryonic stem cell (ESC) derivation and culture. Experience with long-read/long-range sequencing platforms and bioinformatics analysis of complex genomic datasets. Additional Details Normal work schedule is routinely Monday - Friday, from approximately 9:00am - 5:00pm; however, early morning, evening, and occasional weekend work is required by scope of the position. Work is performed in both the laboratory setting with potential risk of exposure to potentially hazardous chemicals and dry ice and in areas where there is potential risk of exposure to zoonotic agents through injury, by direct contact with animals or their tissues and body fluids or by contact with equipment contaminated by animals or their tissues and body fluids.
